Alnylam Pharmaceuticals AKAALNY unveiled its fourth quarter report of 2023, discarding a loss of $1.10 per share. This figure distanced itself from the Zacks Consensus Estimate of a loss of $1.20. Just a year ago, the company experienced a staggering loss of $1.68 per share, eluding last year’s dismal performance. This time around, it seems that the company managed to steer clear of the choppy waters, bringing its losses down to 77 cents per share, marking a considerable improvement from the adjusted loss of $1.39 reported in the previous year’s quarter.

Revenue Recap

Clocking in at a total of $440 million within the quarter, Alnylam’s revenues failed to meet a significant milestone, missing the Zacks Consensus Estimate of $446 million. However, these figures should not be overlooked when considering last year’s total revenues of $335 million, reflecting a substantial increase. Net product revenues proved an influential driving force, amassing $346 million in the quarter, signifying a 32% year-over-year hike on a reported basis and a 30% climb at a constant exchange rate (CER). This incredible feat was underpinned by a swelling patient base for Onpattro (patisiran), Givlaari (givosiran), Oxlumo (lumasiran), and the recently approved Amvuttra (vutrisiran).

Collaboration and Licensing Momentum

The company amassed $76 million in net revenues from collaborators, marking an 8% rise from the $71 million in the year-ago quarter. The primary catalyst behind this surge was the revenues recognized under Alnylam’s licensing agreement with Roche and its collaboration agreement with Novartis, both related to the achievement of specified commercialization and regulatory milestones.

It’s worth noting that in July 2023, Alnylam struck a strategic collaboration with Roche to co-develop and co-commercialize zilebesiran for the treatment of hypertension. The windfall of upfront payment from RHHBY, accompanied by several milestone payments, promises a colossal deal value of up to $2.8 billion. Furthermore, by granting Novartis exclusive and worldwide rights to manufacture and commercialize RNAi therapeutics targeting PCSK9 for the treatment of hypercholesterolemia and other human diseases, including Leqvio (inclisiran), the ongoing collaboration has demonstrated a capacity to further bolster Alnylam’s already sturdy financial position.

In addition to the collaboration with Roche, Alnylam continues to reap revenues from its ongoing collaboration agreement with Regeneron. Together they’re working on ALN-APP to treat early-onset Alzheimer’s disease and cemdisiran, an investigational RNAi therapeutic for the treatment of complement-mediated diseases. The company is entitled to receive license fees from these agreements and recorded royalty revenues of $17 million in the reported quarter, which is a staggering difference from the $2.7 million recorded in the year-ago quarter.

Stock Plunge and Investor Sentiment

Despite these impressive financial figures, the company’s stock suffered a hit, plummeting 10.2% in the last trading session, a direct consequence of the mixed fourth-quarter results. The decline in the stock price also points to investors’ skepticism regarding the changes to the heart disease study’s plan. Over the past year, Alnylam’s shares have nosedived by 33.8%, quite the stark contrast compared to the industry’s decline of 11.5%.

Historical Performance and Future Prospects

Alnylam’s sales performance in the quarter speaks to the sway that the company holds within the pharmaceutical industry. Onpattro (patisiran) and Givlaari (givosiran) recorded sales figures of $79 million and $59 million, respectively, in the fourth quarter, both demonstrating solid year-over-year performance. Despite a 35% drop in Onpattro sales, this was still a commendable beat against the Zacks Consensus Estimate of $75 million. In addition, the initial uptake of Amvuttra was indeed promising, with sales worth $175 million, marking a 156% increase. Moreover, Givlaari managed to achieve an impressive year-over-year increase of 26% in sales.

This robust performance was not a fluke, as Alnylam also recorded a global net product revenue of about $32.7 million for Oxlumo (lumasiran) in the fourth quarter of 2023, reflecting a 37% year-over-year leap. Adjusted research and development (R&D) expenses saw a slight uptick, rising by 3% year over year; meanwhile, adjusted selling, general and administrative (SG&A) expenses declined by 5% year over year. Alnylam managed to maintain a solid financial standing, with its cash, cash equivalents, and marketable securities amounting to $2.4 billion as of December 31, 2023.

A Landscape Beyond 2023

The results from 2023 further demonstrate the strength of Alnylam’s financial backbone, with total revenues reaching $1.83 billion compared with $1.04 billion in 2022. The collective performance throughout the year has visibly contributed to narrowing the company’s losses to $3.52 per share, down from a loss of $9.30 per share reported in 2022, fueled by higher revenues. The figures landed just off the Zacks Consensus Estimate but signify a steadfast trajectory moving forward.
